Mục đích sử dụng

Bộ Kit NKCult (Mã SP: 298) được thiết kế chuyên biệt để phân lập, hoạt hóa và nuôi cấy tăng sinh quần thể Tế bào Diệt tự nhiên (Natural Killer Cells - NK) từ một thể tích nhỏ máu ngoại vi (chỉ 20 mL).

Sản phẩm được định hướng ứng dụng trực tiếp trong lĩnh vực Trị liệu Tế bào Tiên tiến (ATMP). Cung cấp giải pháp toàn diện cho các phòng Lab chuẩn GLP/GMP sản xuất "thuốc tế bào" an toàn, đạt các tiêu chí xuất xưởng lâm sàng khắt khe nhất: Độ sống sót > 90%, độ tinh sạch kiểu hình > 80% (quần thể CD3-CD56+) và hoạt lực ly giải tế bào ung thư mạnh mẽ (> 50%).

Mô tả

Dựa trên triết lý cốt lõi của Y học tái tạo: "Tế bào chính là Thuốc", NKCult không chỉ là một bộ hóa chất, mà là một Hệ sinh thái công nghệ trọn gói (All-in-one), được thiết kế với sự tôn trọng tuyệt đối các đặc tính sinh lý của tế bào NK:

• An toàn tối đa với nền tảng Vô huyết thanh (Serum-free): Nguồn dưỡng chất hoàn toàn không chứa huyết thanh động vật, loại bỏ triệt để rủi ro tạp nhiễm và các phản ứng miễn dịch dị loài.

• Hoạt hóa định hướng không gian 3D: Khởi động quá trình bằng hệ thống vi protein tái tổ hợp phủ bề mặt (Coating Reagent), tạo tín hiệu nhận diện đặc hiệu giúp tế bào bám dính và hình thành cụm tăng sinh tự nhiên mà không cần tế bào nuôi chung (feeder cells).

• Nuôi dưỡng tinh tế theo pha (Phased-Nutrition): Hệ vi chất được thiết kế chuyên biệt cho giai đoạn huấn luyện thụ thể (D0-D3) và giai đoạn bùng nổ sinh khối (D4-D21), đi kèm với các vật tư được tinh chỉnh để bảo vệ tế bào khỏi stress cơ học (shear stress) và sốc nhiệt.

• Chiến lược thu hoạch linh hoạt: Bộ Kit trao quyền quyết định cho nhà lâm sàng với hai kịch bản đỉnh cao:

Mã sản phẩm 298 được định lượng chuẩn xác và đồng bộ cho 1 mẻ nuôi cấy từ 20 mL máu ngoại vi, bao gồm:

Nhóm I: Hóa chất & Môi trường sinh học

• NKCult Basal (298-1): 01 chai (540 mL) - Môi trường nền cung cấp dưỡng chất thiết yếu (Vô huyết thanh).

• NKCult Coating Reagent (298-2): 01 vial (0.5 mL) - Phân tử protein tái tổ hợp dùng để phủ bề mặt mạng lưới kích hoạt.

• NKCult D0-3 Supplement (298-3): 01 vial (2.0 mL) - Hệ vi chất kích hoạt thụ thể giai đoạn sơ cấp.

• NKCult D4-21 Supplement (298-4): 05 vials (8.0 mL/vial) - Hệ vi chất thúc đẩy tăng sinh và mở rộng quy mô.

Nhóm II: Vật tư tiêu hao chuyên biệt

• MNC Collection Tube (298-7): 03 ống (8 mL/ống) - Tích hợp sẵn chất chống đông ACD-A, tối ưu phân lập khối tế bào đơn nhân.

• T-75 Flask Non-Treated (298-5): 01 bình - Bề mặt chuyên dụng cho quá trình phủ protein kích hoạt.

• NKCult Bag 500 mL (298-6): 01 túi - Túi nuôi cấy màng thông khí (gas-permeable) chuyên dụng cho giai đoạn Scale-up.

Điều kiện bảo quản (Storage Conditions)

Sản phẩm chứa các yếu tố tăng trưởng là protein tái tổ hợp rất tinh xảo, BẮT BUỘC lưu trữ nghiêm ngặt theo các vùng nhiệt độ:

• Bảo quản đông sâu (-20°C): Áp dụng cho các vi chất hoạt tính cao (NKCult Coating Reagent, NKCult D0-3 Supplement, NKCult D4-21 Supplement).

• Bảo quản lạnh (2°C - 8°C): Áp dụng cho môi trường nền NKCult Basal.

• Nhiệt độ phòng (Khô ráo, thoáng mát): Áp dụng cho nhóm vật tư nhựa (Bình T-75, Túi NKCult Bag, ống MNC).

[⚠️ ĐIỂM KIỂM SOÁT TỚI HẠN - CCP]: Môi trường tăng sinh D4-21 sau khi đã pha trộn hoàn chỉnh (Basal + Supplement) BẮT BUỘC lưu trữ ở 2°C - 8°C và chỉ giữ được hoạt tính tối đa từ 7 đến 14 ngày. Áp dụng nguyên tắc tối thượng của ATMP: "Pha lượng vừa đủ dùng, dùng đến đâu pha đến đó".

Điều kiện vận chuyển (Shipping Conditions)

Để cấu trúc 3D của protein không bị đứt gãy, bộ kit yêu cầu vận chuyển tuân thủ tuyệt đối quy trình Chuỗi cung ứng lạnh (Cold Chain Logistics):

• Nhóm vi chất cấp đông (-20°C): Phải được vận chuyển ngập trong Đá khô (Dry Ice) bên trong thùng cách nhiệt chuyên dụng, duy trì trạng thái đông cứng xuyên suốt tuyến đường.

• Nhóm môi trường giữ lạnh (2°C - 8°C): Vận chuyển kèm Gel lạnh/Đá lạnh (Ice packs). Yêu cầu có vách ngăn cách nhiệt để môi trường Basal không tiếp xúc trực tiếp với đá lạnh gây sốc nhiệt tĩnh hoặc đóng băng cục bộ.

• Nhóm vật tư tiêu hao: Đóng gói chống va đập ở nhiệt độ môi trường. Đặc biệt cẩn trọng không đè nén làm biến dạng màng thông khí sinh lý của túi nuôi cấy.

• Lưu ý an toàn cơ học: Tuyệt đối tránh lực quăng quật, rơi rớt hay lực xoáy ly tâm làm gãy các cầu nối protein mỏng manh bên trong các vial hóa chất.

The product is intended for direct application in the field of Advanced Therapy Medicinal Products (ATMPs). It provides a comprehensive solution for GLP/GMP-compliant laboratories engaged in the manufacturing of safe “cell-based medicinal products,” meeting stringent clinical release criteria, including: cell viability > 90%, phenotypic purity > 80% (CD3⁻CD56⁺ population), and potent cytotoxic activity against cancer cells (> 50%).

Description

Based on the core principle of regenerative medicine - “Cells are the Medicine” - the NKCult Kit is not merely a reagent kit, but an integrated, all-in-one technological ecosystem designed with strict adherence to the physiological characteristics of Natural Killer (NK) cells:

Maximum safety with a serum-free platform:
The nutrient source is completely free of animal-derived serum, thereby eliminating risks of contamination and xenogeneic immune reactions.

Spatially oriented 3D activation:
The process is initiated using a surface-coated recombinant protein system (Coating Reagent), providing specific recognition signals that promote cell adhesion and the formation of natural proliferative clusters without the use of feeder cells.

Phased nutrition strategy:
A specialized micronutrient system is tailored for the receptor priming phase (Day 0–3) and the rapid expansion phase (Day 4–21). Associated consumables are optimized to protect cells from mechanical (shear) stress and thermal shock.

Storage Conditions

This product contains highly sensitive recombinant protein growth factors and MUST be stored under strictly controlled temperature conditions:

• Refrigerated storage (2°C to 8°C): Applicable to NKCult Basal medium.

• Controlled room temperature (dry, well-ventilated): Applicable to plastic consumables (T-75 flask, NKCult bag, MNC tubes).

[⚠️ Critical Control Point – CCP]:
The fully prepared D4–21 expansion medium (Basal + Supplement) MUST be stored at 2°C to 8°C and maintains optimal activity for 7 to 14 days only. In accordance with ATMP principles: prepare only the quantity required for immediate use.

Shipping Conditions

To preserve the structural integrity of recombinant proteins, the kit MUST be transported in strict compliance with cold chain logistics:

• Frozen components (≤ -20°C): Must be shipped on dry ice within validated insulated containers, maintaining a frozen state throughout transit.

• Refrigerated components (2°C to 8°C): Shipped with ice packs/gel packs, with appropriate thermal barriers to prevent direct contact and avoid thermal shock or localized freezing.

• Consumables: Shipped at ambient temperature with protective, impact-resistant packaging. Particular care must be taken to avoid deformation of the gas-permeable membrane of the culture bag.

Mechanical handling precautions:
